If I remember correctly, though, in Portal 1 the door not only needed the cube to be destroyed, but it also wouldn't open until the timer ran out. Here's an idea: make a laser pointing at a laser catcher, and hide them out of sight. Connect the laser catcher to the door, and the timed button to the laser, and press "start enabled" on the laser. Because of this, the button will deactivate the laser when pressed, meaning that the door won't receive a signal from the catcher, and won't open until the timer runs out.
