You need to go to the left of the Research Center, in the room with the shipping containers. Interact with the thing in the middle to get in one. You'll end up in the central transport hub. Go up the elevator, and you should be able to find your way from there.

Oh, no, you don't have to defeat Chiyou's brother. That is an optional side-quest. For him to be there, you must complete the rest of Chiyou's questline.
