Position #1.This is going to be carry/safelaner. This is the hero that you want to be CS'ing and most of the time its up to the supports to ensure he gets it, which is why they most often go to the 'safe' lane. He has the highest farm priority being that it is usually a hero that needs and/or does really well with farm and items. Any hero can technically be a 1 position (or any position, depending on what your team needs) if he's getting the farm priority of one as determined by his team, which is why sometimes you'll see things like Doom and Centaur safelane even though they are not conventional 1 positions. Examples of 1 positions will be your heroes like Sven, Spectre, Antimage, Medusa, etc. If you've ever played the Dota game before, you know that these are heroes that absolutely need items and farm and will carry your team later and is why they are good 1 positions.

Position #2. Most often the midlaner, 2nd in farm priority only to the carry hero. These are usually heroes that need just a few items to come online, like maybe a blink, a veil, etc, and is generally why they aren't required to get as much farm as the 1. They do however most of the time rely on getting the EXP to match their farm, which is why they usually are left to solo the midlane. Good 2 position heroes would be things like Queen of Pain, Lina, Shadow Fiend, Outworld Devourer, Puck, etc. There are some heroes that become good 2 positions given you have a team that enables them to do so, such as having a mid Medusa if you have a Drow Ranger on your team.(pagod mag english)

Position #3. Usually the offlaner, these heroes rely mostly on just trying to get EXP over farm, which is why they are also given a solo lane most of the time, though usually it is one that they won't be able to rely on getting farm due to the enemy supports usually being there to help their carry. Given that they're lanes are usually pretty hard, heroes most often picked for the role are ones that have a good escape mechanism or are just outright hard to kill. This combination of things usually make heroes like Puck, Tidehunter, Centaur, Timbersaw, etc good offlaners. (NOT Weaver and Necrophos, these heroes need items they will not be able to get from offlaning, I'm looking at you 3k and 4k)(c-p lng naman)

Position #4.This is going to your 'farming' or roaming support, or perhaps even a jungler. These are support heroes that you don't need to have as much farm or experience as your cores, but you'd still like them to be functioning a bit earlier than your 5 and sometimes are even heroes that can transition into being a 4th core. They might be a hero that needs a Blink, but it isn't as important as the first 3 cores getting farm which is why he takes 4th priority. Given that they will generally be heroes that need a bit more than the 5 would, while they can buy wards and other support items, its not always expected of them, at least until they get whatever items or levels necessary to do their job a bit better. Depending on the hero, they will sometimes be the hero that is depended on to ensure all his cores have good laning stages by ganking and roaming. Good heroes for this role can include an Earthshaker, Nightstalker, Sandking, Monkey King, etc.

Position #5. The lowest of farm priority, these are usually the heroes that will be charged with buying most if not all of the support items in the game. They will generally be heroes that don't require much EXP or farm as long as they do their supporting job well of warding and ensuring his carry has a good game. While typical 4 position heroes can also be transitioned to the 5 position, it is much harder to win games if you have 2 typically 5 position heroes. As a rule of thumb, you won't want 2 super squishy heroes that have no ganking or farming potential to be your support duo. This why it might seem hard to win games if your support duo is something like a Lich and a Skywrath. They are both kind of static lane supports which will end up just detracting from the experience of your carry player. These will be the guys that will be pulling and stacking for the cores, and zoning the offlaner when they aren't warding or TP'ing to save their cores. Good hero examples for the 5 position role are things like the mentioned Lich and Skywrath, Crystal Maiden, Bane, Disruptor and others.
