Chose Your Path
Pick a class line to focus on rather than trying to unlock them all. For example I chose to go with the mime - tier 1 to shapeshifter- tier 2 to cartomancer- tier 3. The tier 2 and tier 3 adventurers are much stronger so you'll want to save your money and unlock them asap.

Unlock all tier 1 loot and pick and choose your tier 2 and 3 loot depending on your playstyle. You may want to eventually unlock all the loot but money comes slowly at first. I know all that bling is tempting but try to keep focus.

There are a few blessings to choose from but you can only take one with you into the dungeon. I personally found the crone's blessing to be the most useful. It adds an extra card to your hand which can often make the difference between success and having your face melted off.

I unlocked tier 1 loot and my tier 1 and 2 heros first, then upgraded my blessing to tier 2 and finally tier 2 loot cards. (I haven't made enough coin for tier 3 yet) You may have different priorities but chose a path and stick with it.
Boss Types
The Lazy Boss - This monster will wait in his lair for you to come to him. Don't connect to the boss room too soon or your adventurer may head there before either of you are ready. Take your time and wander the dungeon, laying down monsters, rooms and loot cards to help you level up.

The Aggressive Boss - This monster is on your tail and getting ever closer! You need time to level before it catches you yet it moves two squares each turn. What to do? Lay monster cards in front of the boss to slow it down to one square per turn! Try your best to keep your dungeon path from circling back on itself and tempt your hero forward with monsters and loot so he/she can level.

The Timed Boss - This monster will attack in a certain number of turns. Often the dungeon will also have at least one treasure chest room and a fountain which may give a blessing or a curse. Try to connect to the treasure if you can but focus mainly on laying down those monster cards so you hero will be strong enough when the boss attacks.
Conclusion
I'm still a beginner myself in this game but the above tips have been a big help to me in getting started on the path to greatness. I find playing (and failing!) the best way to learn a game but hopefully this guide will help take away some of the beginning frustrations.

Sorry, I know this little guide is out of date. A lot of new mechanics have been added to the game since I wrote this.

In additon, the "choose your path" advice is good for a dungeoneer, but you should invest better. Do NOT get the curio shop equipment pack, for example, if you're choosing the bruiser/barbarian/knight, as that is really only magic items that won't benefit your character, but will instead cause them to have a lesser chance of getting the things they need, for example.

Timed Boss:
No. You want to lay down a straight path to the boss. Count how many turns (basically, how many squares you can move) until he attacks and see if you can get that chest/fountain, but make getting tot he boss BEFORE he attacks a priority. All timed bosses have a weakness that is only exploitable if you attack them (such as frail - all physical attacks do 1 extra damage).
If you let them attack you, you may or may not be any stronger than if you attack them (based on RNG loot tables, you could get the best equipment early or it could take a while, or you could never get it before the boss attacks). But if you attack first, you WILL get that distinct advantage.

Gonna now reset with the knowledge of these hints, also would it be ok if I were to focus on two?

Thanks for the tip on the 'Aggressive Boss' about laying monsters in his path, I'll have to try that with the minotaur (who has been busting my behind!)

Collect equipment which provides bonuses of the same type. Ability X gives you all the cards from 1 to X in that ability, where higher numbers are stronger. Also, collect ability types, as they add, rather than only the strongest counting.

Those are good tips: it's not clear when you start what the best way to level up is, but sticking to one path and levelling heroes before gear will give you the best results.

Often, the best action to take is none at all: if you're not on one of the timed quests, then don't be afraid to discard the entire hand of cards you're given and skip that turn. Keep doing that until you get some cards you like.

In most dungeons, there is one Tier 1 monster that your chosen hero can beat more easily than others, so concentrate on fighting them as much as possible and take the time to exploit their weaknesses, e.g. build a block of rooms, then put a 'Night Owl' monster in the middle, or lay down a dead end and put a 'Loner' in it: mouse-over the bottom of the cards to get descriptions and tool tips about their strengths and weaknesses.
