Introduction
The wonderful thing about Block N Load is that there is no perfect defense. Any strategy is viable to win a match. There are plenty of useful guides on defending cubes, but sometimes you'll be forced to improvise your own defense, which can be intimidating, but after reading this guide, you'll be comfortable with defending any cube on any map.
Preparation
M.L.G. is a helpful acronym that you can use to remember the three qualities that make up a good defence.

Mobility

You know your base is flawed when the enemy has an easier time navigating your base than you do.
Build jumppads, staircases, and tunnels so that you can get to every inch of your base as quickly as possible. Make it hard for the enemy to infiltrate your base without hindering your mobility. Can the enemy dig through the ceiling? Build a staircase to the roof, so you can kill them before they breach. Is there a bridge to the cube? Take it out, but make sure you can respawn on the other side. Can the enemy spawn directly at your base? Seek out their respawn pad.

iLuminate

On a scale of 1 to 10, how useful are radars when defending a cube? The answer is 11. Make sure your entire base is covered with radars. It's important to see the enemy before they see you. When you know where they're going to breach, you can set up a trap, or stand guard ready to attack when they breach. Setting up radars also lets you see where people set up respawns.

Guard

After you frantically set up your preparations for defence, you need to guard it. In some circumstances the opposition can be very overwhelming and you need a partner to help you guard. Consider how many enemies are attacking, how skilled they are, and what class they are playing as when selecting who to help defend. I find that it's a good idea to have a mix of two class types, for instance, a Nigel (a skill class) and a Tony (a brain class) is a better combination than a Sarge and a Cogwheel (two brawn classes).
Maintenance
P.S.A. is another acronym you can use to remember how you can maintain a lethal defense.

Placing traps
You know how stupid you feel when you step on a mine, or fall into a fake block trap? That's how the enemy feels when they die to one of your traps. The biggest mistake I see players do is put their traps on the rim of the cube, as if they expect the enemy to walk right up to them. Traps need to be placed in areas that the enemy think are safe. For instance, if I see an enemy digging through the wall, I place a mine next to the block I suspect them to breach from. Remember, when they are digging through the wall, they think they are cleverly bypassing your traps.

Setting up turrets
You know what's more awesome than building? Turrets! Players have a tendency to destroy turrets before going for the cube. Thus, turrets buy you and your team more time to destroy the enemy's cube. Be aware that turrets can't shoot directly up or directly down. Place turrets far from each other so they don't get taken out together. Make sure that there's a ceiling above the turret; an unprotected turret can die instantly to Astarella's laser beam, or Cogwheel's mortar strike.

Turrets are expensive, so try and keep them alive with your caulk gun. Healing turrets with the caulk gun increases their damage, just as an FYI. When I play as Tony I prefer to place two turrets at a time, that way it doesn't cost too much to replace a broken turret.

And one last note, don't rely on your turrets to kill everything for you. They are most effective when you attack the enemy that they are aiming at.

Acquiring bricks

I find that most defenses fall apart when the defender becomes starved of bricks. Avoid brick starvation by spending wisely. Determine the value of each block you place. Do I really need two mines to cover this doorway? Does this fake block trap have to be this big? Also, in order to maintain a healthy brick count, dig during a hiatus in combat. Any block that is not necessary for your fortifications is better off being a part of your brick counter. The floor around the cube is a good source of brick income. I'd advise however not to dig too deep, you might end up restricting your mobility.

Also, if your fighting in the front lines, remember that the poor defenders really appreciate it when you pick up supply drops and cut down structures.
As always, human life is the most important
Any defence you build is worthless if you are not alive to guard it. When Astrella launches her laser, or Cogwheel sends in a mortar strike, you should run away. Any turret, mine, tiki that is lost can be replaced in a matter of 2-3 seconds. It takes 10-30 to bring yourself back. Health and Ammo blocks help you stay alive. Save health pickups for when you need them. If you have to, call over a friend to provide some assistance or heal you.
Conclusion
I hope I was helpful, and that you've become a better player thanks to this guide. In the comments I'd like to hear about your crazy, improvised defenses. Any feedback is greatly appreciated. Thanks, and don't you ever....

Helped me a lot in my playtime as a Defensive. Many Offensives never care about their team in the back an suicide mission in.
