If I could direct you towards the Discussion threads for the game, you may find a solution to your issue there. Otherwise, try going to the Kumite/Sparring section of the game and practicing - Some of the special combinations must be input quickly while some require a different rhythm.

But to answer your question, there are two ways to defend in the game:
- By simply pressing up or down your character will "soft block", i.e you'll see a small block spark.
- If you press both punch buttons (X+Y on controller or U+I on keyboard) you can do a "hard block" which is basically a parry. The game will freeze for a second and pressing any button will result in a free kick counter.
I personally don't find the "hard block" to pay off very much other than to give you a break to regen your Ki.
