Overview

1. Get to the tower's second floor, Dave's Home

2. Find and destroy the statue

3. Enter the portal, defeat the boss

2. Find and destroy the statue

Get to this part of the map and destroy this statue, a hidden path will open.

3. Enter the portal, defeat the boss

After entering the portal, a boss fight will start, no screenshots in this section because I don't want to spoil it.

4. Done

After defeating the boss, Cleopatra will be unlocked. You can continue the level normally or go straight back to the base to put her on your team.

Cleopatra is part of the base game. And as far as I know, all other characters are too. There's no additional DLC for the game right now, I think when the devs say "DLC" in the update news for the game, they just mean the new content added to the game.

I keep seeing reference to a DLC, but cannot find a DLC for this game. Do you need to install anything additional, or is this part of the base game?
