Since a 50+ Chain could be really difficult to achieve ingame (Board is 63 spaces + your character), here are some tips to get it :

Level : obviously go to the Plains and try to stack as many Beetos as you can
Character : I will suggest first Mole Knight for its ability to swap positions but you can use Treasure Knight or Shovel Knight if you please. Scrap Knight is another good choice.
Items : in fact, items such as Speer or Bombs are really at your disadvantage but you can remove them in the options :
Last but not least, Speed : to do it more doable, you can slower the speed to 0.25, I suggest to remove it totally only with Mole Knight or Scrap Knight. With any other characters, you can be blocked at the very end of the level (see screenshot with Treasure Knight) and want to wait for some extra Beetos. It will also be more easier with Green Blorbs :
