Introduction
You want to hunt Fluffies, you and your friends are kitted out in the finest Howell-Barrex equipment, you have the weapons to survive STROL domination of Mars, and you got the teamwork of an entire Condatis Platoon with the ingenuity of Insurgents.

Know that the F.L.U.F.F.Y Hunt is not for fresh recruits, disorganized teams, moderately experienced veterans, teams who don't know they're doing the F.L.U.F.F.Y Hunt, players who are not equipped for F.L.U.F.F.Y Hunting, and in general anything but highly experienced players with tight coordination.
Hunting Grounds
Now onto the meat of this guide. I only know of how to fully initiate the F.L.U.F.F.Y Hunt on a select few maps, Prevent Infestation Spread and Noachis Colony, although I do know of how to trigger it generally on most maps.

There are several map requirements for initiating the hunt that I know of. They are
    1. Difficult is set to Normal at the minimum
    2. Total difficulty is set to 3 "Skulls"

If these settings are not initialized, then this guide will not help you in initiating the hunt, whether by my lack of knowledge or it flat out not letting you.

Now onto more specifics, you will need to do two things. The first is to trigger the "World" power relay. This power relay is on the edges of the map, unlinked to any prefabs. This means they will always be in the same spot on the map.

The second is to find the briefcase. These are easily distinguished by being a briefcase, usually found on a road. They are also not linked to any prefabs to my knowledge, but it is not known for sure. The briefcase must be activated before the third wave on any map, otherwise F.L.U.F.F.Y Hunt will not trigger.
Finding Prey (Prevent Infestation Spread)
So, now you got your kit polished, your team is prepped, now you just need to start it. Know that F.L.U.F.F.Y Hunt has to be initiated in the first three waves I believe, or else it will not trigger.

There is a major thing to know about initiating F.L.U.F.F.Y Hunts, that I know of at least.

THE FIRST POWER TERMINAL HAS TO BE THE WORLD TERMINAL

Note that this is not a fully hard rule, but it is a good rule of thumb. For example, you can maybe activate one power terminal before the world terminal, and maybe activate two, but definitely not more than five. It is still important to let your team know you are doing a F.L.U.F.F.Y Hunt however.

Standard item containers, basic containers and such, are perfectly fine to use to my knowledge however. They will not cause the initialization of the F.L.U.F.F.Y Hunt to fail.

The Power Terminal that triggers F.L.U.F.F.Y Hunt on Prevent Infestation Spread is to the North-East of the map, and only one person should activate it. It must be activated before wave three, otherwise it will not activate the console. (Thanks to E.D. for this part!)

Power Terminal in question.

Power Terminal location on Map.

The specific terminal, if a prefab spawn has another power terminal near it, is always the one closest to the road.

Now that you got the power on, you need to find the console. Fortunately, it is relatively consistent in its spawns on this map. It is always on the center of the road to the right of the central prefab structure.

Where the Console usually is.

Brief-case looking thing in the center is the console.

If you have followed the instructions properly, then that console should be lit up and when hovered over display a string of 4 sets of two numbers. Activate it, and it will trigger the initiation sequence for F.L.U.F.F.Y Hunt. Note that it will take a little while to actually add in the secondary objective, and it might be based on the team being clustered.
Finding Prey (Noachis Colony)
Welcome to Noachis Hunter! Make sure your boots are on tight because you'll be running far through the heaps of the dead.

Thankfully, there will not always be a tree in front. It will always be in that corner however.

Note its closeness to a Teleporter Pad, and to a wall outcrop.

Now that you got the power on, you need to find the console. Thankfully, user Darc24 has found it. It is always near the teleporter pad near the northern tip of what I will call the Arboreal Preserve.

Note the nearby Teleporter pad.

The Hunt Begins
So, assuming everything went well, you activated Project TERRA, and the hunt is on! Now you just need to go down swinging.

Waves:
Initially you'll be fighting off fist-fulls of F.L.U.F.F.Ys and other car-sized dogs, and then have to take down a Specimen entering the field before you can be said to have fully completed the Hunt.

Thankfully, all but the Specimen it can be relatively easily killed if everyone keeps to a specific place, and are willing to wait several waves.

Strategy (Howell Box):

Works only on lower difficulties, as the damage boost provided by higher levels is too devastating for a single combat engineer. Double engineer might work but it's a heavy specialization.

No artillery is suggested for this strategy, as it relies upon having a fully blocked off room with no holes, so then no monsters can slip in.

You'll want a room with a Blast Door as the only entrance to hold the line. The prefabs Elysium HQ and Small Bunker are one of the best ones, as for Elysium HQ there is a Zipline near the highest room, which you'll want to take, while for the Small Bunker it has an entrance easily trapped to kill the horde.

For the waves, possibly the best option is to bunker down in a room and not get your face eaten by the horde. Unfortunately you cannot get that door to last longer if you do not have a door blocker, nor can you passively kill the F.L.U.F.F.Ys with items.

Thankfully, that's why Trinity gave us Combat Engineers.

With the usage of Fortify & Rebuild, Electric Barrier, and Repair, a single Combat Engineer at a stretch can handily handle the entirety of the horde, and racking up enough kills to get the medal. The Electric Barriers are the more important part of the build, as they are the passive damage source for the waves. Fortify & Rebuild is used to give your door a bit extra life and deal a bit of damage to them as they attack it, while Repair is spammed near the door to make sure that the door lives. A door blocker item is useful, but not required by this strategy.

Note that the F.L.U.F.F.Ys and other dogs can sometimes still attack you through the door if you stand right next to it, so be careful Engineer!

After a couple of minutes, all of the F.L.U.F.F.Ys should be dead by now, and thus you can emerge onto the field and kill the Specimen at your relative leisure.

Thank you for telling me that, I will update the guide accordingly.

I've actually gotten Fluffy Hunt to trigger on Normal difficulty. 3 skulls still required though, and couldn't trigger it on Easy. It saved me a lot of time when I was going after 100 Fluffy kills.

Thank you, forgot to add in that the briefcase must be activated before Wave 3.

The briefcase is almost always found on the center road network like in your Prevent Infest Spread map.
You also have to do it before wave 3 otherwise it won't spawn.
