Having problems lining up headshots my dear spy? Fear not, for I will mentor and train you to be the ultimate ambassador sharpshooter (note that there was exaggeration in that statement). Seriously though, this is a simple and effective guide for any skill level spy that wants to improve upon their ambassador capabilities.

Aiming
For the ambassador, there are many components of getting a successful headshot, including a 1-1.5 second cooldown rate, general aiming, and even crosshair preference.

The cooldown rate balances out the ambassador because it prevents constant headshot spam. After you shoot the first bullet, this cooldown will begin. If you fire before the cooldown finishes, your shot will not COUNT as a headshot, and the bullet will be less accurate. However, if you shoot any time after the cooldown rate finishes, you can get a headshot, and the bullet will generally be very accurate.

There are many tips for lining up headshots like not moving your mouse as much, but simply moving left and right to line up the shot. If your crosshair is at the appropriate height, you can press A or D to position your crosshair onto a target's head. If this doesn't work after giving it a good try, you can lower your mouse sensitivity.

Believe it or not, your crosshair could be the problem. You should try all the default crosshairs, and see which one(s) work for you. Also, changing the color to a more noticeable one can improve your headshot ratio.

Personally, my crosshair is a bright teal hollow circle. I use that specific crosshair because all I have to do is put the target's head inside of it.

Lastly for crosshairs, you can change the size. When you pull the trigger of the ambassador, your crosshair becomes larger and slowly shrinks back down to normal size. If you don't want a giant thing popping out of your screen, make the crosshair smaller.
Prediction
Not all targets will be moving in a straight line towards you, like rocket-jumping soldiers or pesky scouts that are attempting to dodge. For these victims, you will need to predict where there head will be when your bullet is able to hit. It may sound difficult, but as always, you just have to practice.

For rocket-jumping soldiers, the easiest time to shoot them is right before they begin their descent. There is a second or two where that soldier is virtually motionless, which is when you should attempt the headshot.

As for double-jumping scouts, you should generally wait until they jump a second time so they can't suddenly change direction (unless they have an item that grants extra jumps). After their second jump, you should aim a tiny bit before their head. For example, if the scout jumps right, aim barely to the right of his head.
Effectiveness
A lot of spies use the ambassador, but don't fully understand its capabilities. A headshot on a medic can force him to use his uber early, giving your team a HUGE advantage. Also, it can quickly disable any class in the game.
