1. Jumping from side wall to hidden place with Spider Rune

At first, you have to jump from left side wall to this hidden place. You need the Spider Rune here.
2. Using the Homunculus Rune to bring blueprint to you

When you are already in hidden place, you just click "V" on keyboard and jump with blob to the blueprint. If you step on it, it actually sticks to you. Then just click "V" again, to acquire the blueprint.
3. Well done, you just got new mutation

Yeah, bravo.

Disengagement - If your health falls under 20% of maximum, you become fully protected for 6 seconds. Can only occur once every 120 seconds.

You don't need the spider rune to get that blueprint actually.
Position yourself left of the torch and door (ground level, near the starting position) and use the homonculus from there.
It'll stretch to its max, but you'll be able to grab the item.
