Introduction
Here's a detailed list of every achievement in the game and the method(s) I went to obtaining them. The game itself is relatively easy to complete with the proper methods, but keep in mind that there are co-op achievements so you'll need a partner if you plan on completing everything.

Simply hit Ctrl+F and type in the achievement name for a quick look at how to get that particular achievement. If you have any further questions or need additional help, feel free to post in the comments below and I'll address them the best that I can. Happy hunting!

Happy hunting!

Enemy Achievements
First Blood
Kill 1 alien.
Kill an alien. Simple, right? You'll earn this one immediately during the prologue.

Eradicator
Kill 100 aliens.
Kill 100 aliens. You'll earn this over the course of the campaign. No special grinding needed.

Exterminator
Kill 1,000 aliens.
Kill 1,000 aliens. You'll earn this over the course of the campaign. No special grinding needed.

Annihilator
Kill 10,000 aliens.
Kill 10,000 aliens. You'll earn this over the course of the campaign. No special grinding needed.

Xenocide
Kill 50,000 aliens.
Kill 50,000 aliens. You won't kill this many during the course of the campaign. You will need to grind for this one, but do so indirectly. There is another achievement "Master Strategist" that requires you to earn 100 gold medals. It is recommended to grind that one out and this one will come with it naturally. Do not replay missions you already have a gold medal on otherwise you'll just be creating more work for yourself.
Power Core Achievements
Hey! That's mine!
Kill an alien carrying a power core.
You'll earn this one over the course of the campaign. Once an alien takes a power core and you kill it, this achievement will unlock.

Happy Returns
Recover a loose core.
You'll earn this one over the course of the campaign. As mentioned in the above achievement, once that loose core returns to the reactor, you'll unlock this achievement.

Close Call
Save a core that is less than 10 seconds away from the map's exit point.
You may get this one without trying, but if you're struggling to complete it an easy way to do it is to build one tower, sell it (this triggers the enemies spawn timer) and wait for the aliens to collect cores and head towards the exit. Once they are towards the end, use your orbital laser (General Cai) to instantly kill them - finish the level to unlock this achievement.

If you're unsure when the ten second marker is at, right click the alien to see their "exit time" here:

Untouchable
Beat a Chapter 4 mission or higher with zero cores captured.
You'll likely get this one naturally as you play the campaign. Aliens can pick up the cores without effecting this achievement as long as they don't escape with them.

Resource & Boss Achievements
Surplus
Win a mission with at least 1,000 resources remaining.
Complete any mission with 1,000 or more resource credits remaining. It is optional to sell any leftover towers/boosters at the end of the last wave and it will count towards your total; you have a limited window during the end of each campaign mission while the dialogue carries on before the score sheet pops up and ends the level.

Penny Pincher
Win a mission with at least 5,000 resources remaining.
Same as above, but this time with 5,000 resource credits remianing. If you're having difficulties with this you can re-play any mission in "practice mode", which generates 25 resources each instead of the standard 10 in the main campaign.

Salvage Rights
Kill an alien boss creature. (Juggernaut, Rumbler, Turtle, or Crasher)
Defense Grid 2 doesn't have traditional boss types, but the above mentioned are the toughest enemies you'll fight. Killing any one of these types of enemies will unlock this achievement. You'll start encountering these enemy types about a third of the way into the campaign.

Filthy Rich
Win a mission with at least 10,000 resources remaining.
The same as above, but this time with 10,000 resources. This can be difficult during the campaign so it is recommended you play a practice mode or grinder mode to obtain this achievement.
Medal Achievements
Thanks For Playing!
Earn Bronze on a Single Player mission.
You'll easily earn this achievement by playing the game normally. Note, this achievement does not unlock when playing co-op.

Field Promotion
Earn a silver (or better) medal.
Just as the bronze medal above, earning a silver (or gold) will unlock this achievement. Note, while it does not say single player like the bronze medal, this achievement can only be unlocked in single player and will not unlock in co-op mode.

Flawless Victory
Earn a Gold Medal.
As mentioned above, you will need to earn a gold medal in single player for this to unlock. Gold medal any one level to obtain this achievement.

Alien Tears
Earn a gold medal on every mission in Story mode.
This one is easier than it sounds. You'll probably earn most of them without trying, but if you're having difficulties, augment your towers with a few "score boosters" in your tower's kill zone(s) to help inflate your score during play.

The Path Most Traveled
Win a Gold medal on mission 19 without using the Command Shuttle.
Like most missions, you can buy extra tiles from your command ship. However, in level 19 you must complete the mission without using your shuttle to earn this achievement.

Planet Defender
Earn a silver medal (or better) on all missions in the Story mode
A very straight-forward achievement - you'll earn this as you progress the story within the first few missions of the game.

Master Strategist
Earn 100 Gold Medals on any missions.
This will likely be the last achievement you have to earn because it involves replaying levels multiple times and earning gold medals to unlock. There are also a few rules that the game does not tell you so let's have a look. Here I'm looking at level two:
First things first, you must earn all 100 medals on the same difficulty. While the achievement does not specifiy that, earning 50 on easy and 50 on normal will not unlock it. You need to earn all 100 gold medals on the same difficulty.

Secondly, the game only recognizes single player gold medals (co-op campaign still counts) so any "DG Coordinated Defense" medals you earn will not count towards your gold medal count. Remember, if you earned a gold medal on every mission of the campaign, that is already 20 towards your total, so stick to the easy modes like "Grinder, Super Grinder, Fully Loaded, Focal Point, Practice Mode and even Limited Towers/Resources". I wouldn't recommend "Power Outage" unless you want a challenge.
Multiplayer & Reload Achievements
Brothers in Arms
Earn Bronze in a Co-Op Multiplayer mission.
This achievement requires a second player to earn. During the co-op mission you must earn at least enough points to merit a bronze medal. The points are not shared like the normal co-op compaign so if your partner earns the medal but you don't make enough points, you will not earn this achievement.

Note: there is no achievement for a silver medal - only bronze and gold.

Go Team!
Earn Gold in a Co-Op Multiplayer mission.
This achievement requires a second player to earn. During the co-op mission you must earn at least enough points to merit a gold medal. The points are not shared like the normal co-op compaign so if your partner earns the medal but you don't make enough points, you will not earn this achievement.

Note: there is no achievement for a silver medal - only bronze and gold.

Warning Shots
Play a Competitive Multiplayer mission.
This achievement requires a second player, but in a competitive game. You do not have to win a match, just play one to earn this achievement.

Winning Shots
Earn a Victory in a DG Fighter Multiplayer mission.
This achievement requires a second player, but in a competitive game. You do have to win a match, just play one to earn this achievement. This achievement can only be unlocked in "DG Fighter" mode, located here under the multiplayer option:

Retry
Use the Reload Checkpoint option.
This option is only available in single player, not co-op. Anytime during a mission hit the default "backspace" button to reload the current level to a previous point in the game. At the end of the mission this achievement will unlock.

If At First You Don't Succeed, Retry Again
Use the reload checkpoint option five times in a single mission.
Much like the above achievement, but reload five times in one mission. Remember, the reload button by default is "backspace".
Building & Upgrade Achievements
Leadhead, Pew Pew, The Not-So-Friendly Skies, Burn Baby Burn, Great Ball of Fire, Not So Fast, Head Trauma, High Voltage, Kaboom!, Would You Like a Boost With That?
Build 100 of "type" towers
You might unlock a few of these during your campaign run; if you're missing any, you'll earn them as you grind out the 100 gold medals achievement.

Full House
Build and fully upgrade each tower type in one mission.
In one mission, build one of every tower-type and upgrade it to level three for this to unlock.

Tower Expert
Build every tower and every upgrade for each tower.
The same as the "Full House" achievement, but you don't need to upgrade them to level three; building them all and upgrading them to at least level two will unlock this achievement.

Now With Sprinkles
Enable a Tower Augmentation item on any tower.
Tower augmentations are the boosts you can give your tower if you build them on a boost module. Select any of the three to unlock this achievement.

10 Items No Less
Enable a Tower Augmentation Item on 10 Towers.
Exactly like the achievement "Now With Sprinkles", but do this with ten towers in total.

Death From Above, More Please, Pumped Up Towers, No Fast Blast, Shoot That Guy, Go To Your Home
Use the...Special Weapon ("Commander's Name Here").
These achievements are unlocked when using a commander's specific ability. Simply using it in a mission and then completing it will unlock it.

Gun Crazy, Shell-Shocked, Firebug
Using only [Gun, Cannon or Inferno Towers], Win any chapter 4 (or later) mission.
These sound harder than they really are. Since you're only trying to win the mission, score and other objectives don't matter. If you're having trouble earning this, play them on "Practice Mode" for the extra money or "Focal Point" for the damage boost from your cursor and spam towers in every possible building location you can.

Indecisive
Sell 10 towers or more in a single mission.
Build and sell any ten towers in a single mission to unlock this achievement upon completion of the level.
Building & Upgrade Achievements
Yellow Beats Green
Upgrade a level 1 tower to a level 2 tower.
Upgrade any tower and you'll earn this achievement.

Minimalist
Using only level 1 towers, Win any Chapter 4 (or later) mission.
This one can be a little tricky. My personal preference is to do it on "Mission #14 - Surface Tension" with this setup that I'll explain. Click on it for a larger view:
For this, I'd recommend taking Commander Cai's orbital laser to avoid leaking any cores. Stage one you'll want to build a row of concussion towers and staggered temporals to slow the enemy down. I always equip my temporals with disruptors for enemy shields, but feel free to put them where you please. Funnel them far right because the spiral walkways will come up midway through the level.

When the walkways appear I always throw cannons equiped with their slow ability in the central square and then up top pack the center with temporals and meteors and/or towers as well. Then onto stage three I lastly funnel the enemy through more concussion towers.

Since this level has multiple boss enemies, I save the orbital laser for those since level one towers have a hard time putting ones like the Juggarnaut down effectively. With enough luck you can both gold medal the level and earn this achievement. Good luck!

Full Potential
Win a chapter 4 (or later) mission with only level 3 towers only and no selling.
Fairly straight forward to complete - pick any mission from chapter 4 onward and before the end of the level, upgrade all towers to level three without selling anything.

No Sale
Win any mission without selling any towers.
Simple. Play any level without selling a single tower.

Liquidator
Sell 10 towers.
Selling ten towers over the course of your time playing will unlock this achievement. You do not have to sell ten towers in one mission - this achievement is cumulative.

Diversity
Build 1 of each tower type in any single mission.
A self-explanatory one - build one of every available tower during any mission to unlock this achievement.

Arsenal
Build 20 towers of any type in one mission.
Build any 20 towers on any one map to unlock this achievement.

Full Defense
Build any 50 towers of any type in one mission.
Pick any one map and build 50 towers on it. You can either obtain this during the main campaign or easily during the "practice mode".
Miscellaneous Achievements
Boot Camp
Finish all the missions in Chapter 1 in Story Mode.
Simple enough - finish the first chapter of story mode for this achievement to unlock.

Base Defender
Complete the single player campaign in Story Mode.
Complete all 20 missions, regardless of medal level, to unlock this achievement.

Confident
Use the speed up control at 3x for a total of 60 seconds in any mission.
You can toggle the speed by the default "F" key in game. During any one mission, have the speed at 3x for a minute to unlock this achievement. Note, you can toggle it on and off as the timer is cumulative.

What IS that?
Inspect 10 aliens.
Right-click on any ten aliens in total and bring up their data to unlock this achievement.

Siege Breaker, Master Siege Breaker
Survive 100 waves on Grinder/Super Grinder challenge mode on any mission.
Not hard, just time consuming. From level three onward you'll have the option to do grinder modes, which put you against 100 waves of enemies. Grinder involves waves of walkers and such while super grinder contains every enemy in the game, making it the more difficult of the two.

That being said, you're just striving to win, so spam towers and augmentations and you should be able to complete them without much trouble.
