Newt Scamander arrives in New York City in 1926 to bring a Thunderbird to Arizona, but a mix-up with a non-magic man named Jacob Kowalski causes some magical creatures to be set loose upon the city.

With the help of sisters Tina and Queenie Goldstein, the guys track down the creatures but also arouse suspicion of the MACUSA and Auror Percival Graves.

A dark magic force called an Obscurial is set loose as a result of a magic child suppressing their powers. The child turns out to be an abused young man named Credence Barebone, who holds the most powerful Obscurial. Although Newt tries to save him, the MACUSA Aurors kill Credence. Graves is revealed to be Gellert Grindelwald, the most powerful dark wizard of that age, pre-Voldemort.

Jacob's mind must be erased to avoid exposure. Newt gives him Occamy eggshells to start up his own bakery, and he is later visited by Queenie.
