Does the AI always use the default leader or can i change/choose it in single player?

There an option to force default leader. So I think normally it's random.

I hope with more leaders, there will be soon an option with an update or so. For example Ptolemaios starts in egypt and not greece. Would be nice to choose it.
on the old world map, choosing Ptolemy should start you in Egypt, unless they changed it

If you start a Hotseat game with only 1 Human player, you can select the AI nations and leaders.

Could you then switch to letting the AI play that other nation?

On another thread I'm wondering why I always research the most techs yet the AI's consistently are ahead of me in many techs. Do AI's research the free stuff or do they get them for free? Do AI's only end up paying to research the basic techs on a script?

A Hotseat game with 1 Human player is the same as a single player game, if you allocate all the other players as AI then yes the AI will play them.

If you want the AI to take over for the Human player as well, you can use the console command 'autoplay x' where x is the number of turns to play.

The AI generally does value the bonus techs. It doesn't get them for free. Outside of the AI starting settings, which can give the AI a few free techs at the start of the game, the AI plays by the same rules as the player in almost all respects, including tech research.

Thanks Fluffster for the informative answer. I wish I'd known about the autoplay feature when first starting just to see the game in action. However I'd never want an AI to play my side in any fashion. I'll also have to try the hotseat mode, glad that's a feature in this game.
