This is not something I've seen before personally. Are you perhaps able to show a small video, so I can see what you're referring to?
If you haven't done so already, tried enabling V-Sync. It can help with older titles when mechanics don't work correctly.

- Playing on 144Hz -> on the final jump from the ledge onto the bar just in front of the cage with the mechanism to lift the key the jump itself will fail, as instead of grabbing the bar Ezio will hit it with his legs
- Playing on 60Hz -> same procedure, but now Ezio finally grabs the bar
