I think there's a good chance of a sequel. There are a lot of eggs in the ending, and the game was more successful than expected.

Well a sequil is hinted at in the ending of the game. The game made a ton of money so it's now a successful franchise. So there is a lot of reason for a sequil. But the main factor here is developer interest. Two Star currently seems to be working on his Cuffbust project. So maybe he's gonna decide to make a Choo Choo charls 2 after Cuffbust. But at the end of the day it's really up to him wether we get a sequil or not.
