As you clear levels, that total time will decrease (ex: You clear a level with a time of 30.00s, then the total decrease to 379:27.41 - (99.99 - 69.99) = 309.28.41).
The speedrun achivement is simply to get a total time low enough. No need to speedrun the whole game in one go. Just speedrun each stage one by one until you get S-Rank on almost every stage (there's a margin, so getting A on some stage is ok). I'd recommend skipping 1-1, as that stage S-Rank is completely ridiculous and requires near perfection to get (took me over 100 tries).
Earlier stages tend to be a bit harder, since they are shorter and have less element to interact with (the margin of error in therefor very small). In later stage, the margin of error is bigger, so you don't have to make perfect jump every time.
Have fun figuring out the "speedrun" strategy of each level (it's a fun puzzle)
